| Description | Purity >95%. The positive transcription elongation factor (P-TEFb) is a protein kinase composed of cyclin-dependent kinase (Cdk) 9 and a cyclin subunit. P-TEFb controls the elongation phase of transcription by RNA polymerase II. It is also a cellular co-factor required for activation of transcription of the human immunodeficiency virus (HIV)-1 genome by the viral transactivator Tat. Flavopiridol is a potent, orally available inhibitor of CDKs. It most effectively blocks the Cdk9 activity of P-TEFb (IC50 = 8 nM), which in turn can prevent transcription or HIV-1 replication. Flavopiridol inhibits all other CDK isoforms (IC50 = 100-300 nM). It less effectively inhibits epidermal growth factor receptor and protein kinase A (IC50 = 21 and 122 µM, respectively). It has potential applications in cancer therapy, particularly in combination with other modulators. Flavopiridol also suppresses synovial hyperplasia and joint destruction in collagen-induced arthritis in mice. |
