| Description | Purity >98%. BCX4430 is an adenosine nucleoside analog with broad-spectrum antiviral activity with EC50 values ranging from 3.4 to >100 µM against a panel of Ebola, Marburg, bunya-, arena-, paramyxo-, corona- and flaviviruses. It undergoes anabolism to BCX4430-triphosphate in Huh-7 cells followed by pyrophosphate cleavage to BCX4430-monophosphate, which is incorporated into RNA strands to induce non-obligate RNA chain termination. BCX4430 inhibits Ebola virus minigenome RNA replication and hepatitis C virus (HCV) NS5B RNA polymerase activity in a dose-dependent manner. In vivo, BCX4430 (5-150 mg/kg) increases survival in mice infected with Rift Valley fever virus. It increases survival of Marburg virus-Musoke infected guinea pigs and cynomolgus macaques when administered pre- or post-infection. BCX4430 (300 mg/kg) also protects mice from mortality in a mouse model of severe Zika virus infection. |
